چکیده
Mitochondria from the livers of normal rats and L-thyroxine (LTa)-injected rats (5 fig/g, 2 hr before killing) were studied by rate tonal centrifugation to detect early changes in physical properties and in composition. LT, t.reatment causes the fractions containing intact liver mitochondria (as judged by succinoxidase activity) to significantly shift their distribution to the heavier end of a linear sucrose gradient, indicating an increase in sedimentation rate. Iodine distribution follows this shift in mitochondria. Iodine concentration increases 26-fold in mitochondria from LT1-treated rats. In addition, iodine concentrates in the less dense part of the gradient in the LTItreated group. Submitochondrial particles from the livers of injected rats were prepared by 15 min sonication and resolved by isopycnic zonal centrifugation. Distributions of iodine, cytochrome aa3, RNA and monoamine oxidase activity indicate that LT4 is associated with the inner membrane of mitochondria.

Fractionation of lodinated Particles and Mitochondria from Thyroid by Zonal Centrifugation and a Study of their Heterogeneity By RAYMOND MIQUELIS and CLAUDE SIMON
1. The subcellular particles of horse and rat thyroids were fractionated in a B XIV zonal rotor on a non-linear gradient of Ficoll after labelling with radioactive iodine in vitro (horse) or in vivo (rat). In the horse, the resulting fractions were analysed for radioactive iodine, protein and enzymes representative of certain subcellular particles. In the rat, iodine turnover and thyrotrophin s...
متن کاملMaintenance of respiratory control in mitochondria after rate zonal centrifugation.
The respiratory control of rat liver mitochondria is lost when they are subjected to rate zonal centrifu~ation in a sucrose gradient (8.0% to 46.6%, w/w) at values for r t necessary for resolution. High sucrose concentrat ion and high ~ 2 t are bo th responsible. Respiratory control can be maintained in iso-osmotic Ficoll + 8.3% sucrose media, and after zonal centrifugation in such media at val...
متن کاملDifferential action of iodine on mitochondria from human tumoral- and extra-tumoral tissue in inducing the release of apoptogenic proteins.
Iodide is actively concentrated in the thyroid gland for thyroid hormone biosynthesis. Excess iodine has been observed to induce apoptosis in thyrocytes and mammary cells. The mechanism of iodine induced apoptosis is poorly understood. Among various cell organelles, mitochondria is known to provide conducive environment for the organification of iodine, i.e. iodination of different proteins. Mi...
متن کاملIsolation of Relaxing Particles from Rat Skeletal Muscles in Zonal Centrifuges
Supernatants of rat skeletal muscle homogenates were fractionated by differential centrifugation and by zonal centrifugation in sucrose density gradients. Cytochrome oxidase was employed as an enzymatic marker for locating mitochondria. The subcellular fractions were also assayed for their ability to prevent the ATP-induced contraction of myofibrils. Both the mitochondrial and microsomal fracti...
متن کاملSTUDIES ON ISOLATED CELL COMPONENTS XVII. The Distribution of Cytochrome Oxidase Activity in Rat Liver Brei Fractionated in the Zonal Ultracentrifuge
The zonal ultracentrifuge was used to separate the subcellular components of rat liver brei into soluble phase, microsomal, mitochondrial, membranous fragments, and nuclear fractions during a single centrifugafion. The centrifuge was run at 10,000 to 30,000 RPM for 15 to 240 minutes, and the rotor contained a 1200 ml sucrose gradient, varying linearly with radius from 17 to 55 per cent sucrose ...
